A new compound 3-(hydroxymethyl)-2,2?-biphenol (HMBP) was synthesized. Complexes of this compound with three N-bases were studied by FTIR and 1H NMR spectroscopy. In the 1:1 mixture of HMBP and triethylamine (TEA) in chloroform, the complexes are formed completely. The hydrogen-bonded chain of these complexes shows large proton polarizability due to collective proton fluctuation. In acetonitrile the alcoholic group of HMBP is no longer bonded to the hydrogen-bonded chain. The rest of the hydrogenbonded chain still shows proton polarizability. In the 1:1 mixtures of HMBP with a stronger base 7-methyl-1,5,7-triazabicyclo[4.4.0]dec-5-ene (MTBD) the phenolic proton transfers to MTBD and is localized there. In chloroform the protonated MTBD is weakly bonded to HMBP via two other hydrogen bonds. In acetonitrile the complex dissociates. The complex between HMBP and urotropine is not formed completely and all hydrogen bonds within this complex are asymmetrical and weak.

A small collection of C2-symmetry hydroxylated biphenyls was prepared by straightforward methods and the capability to act as inhibitors of tyrosinase has been evaluated by both spectrophotometric and electrochemical assays. Our attention was focused on the diphenolase activity of this enzyme characterized by the absence of the characteristic lag time of enzymatic reaction of its monophenolase activity. To this purpose, we evaluated the capability of tyrosinase to oxidize a natural o-diphenol substrate to o-quinone analyzing the changes in the UV?Vis spectrum of a solution of caffeic acid and the reduction of the cathodic current in a tyrosinase-biosensor, respectively. Results of both the methods were comparable. Most of the compounds possessed higher inhibitory activity compared to compound 1, a known hydroxylated biphenyl inhibitor of tyrosinase.

Human immunodeficiency virus (HIV) reverse transcriptase (RT) associated ribonuclease H (RNase H) remains an unvalidated antiviral target. A major challenge of specifically targeting HIV RNase H arises from the general lack of selectivity over RT polymerase (pol) and integrase (IN) strand transfer (ST) inhibitions. We report herein the synthesis and biochemical evaluations of three novel 3-hydroxypyrimidine-2,4-dione (HPD) subtypes carefully designed to achieve selective RNase H inhibition. Biochemical studies showed the two subtypes with an N-1 methyl group (9 and 10) inhibited RNase H in low micromolar range without siginificantly inhibiting RT polymerase, whereas the N-1 unsubstituted subtype 11 inhibited RNase H in submicromolar range and RT polymerase in low micromolar range. Subtype 11 also exhibited substantially reduced inhibition in the HIV-1 INST assay and no significant cytotoxicity in the cell viability assay, suggesting that it may be amenable to further structure-activity relationship (SAR) for identifying RNase H inhibitors with antiviral activity.

In the middle of this ring is a dianionic oxygen atom placed, coordinating to the three L2Zr centres in a planar mu3-coordination (bond angles 120,05). Five lithium ions stabilize the aniomc backbone by bridging the biphenolato chelate ligands, which form seven-membered chelate rings with the atoms. 1H-, 13C-, and 7Li-NMR spectra exhibit that the solid state structure remains unchanged in solution (thf).

Honokiol is a natural product and an emerging drug for a wide variety of malignancies, including hematopoietic malignancies, sarcomas, and common epithelial tumors. The broad range of activity of honokiol against numerous malignancies with diverse genetic backgrounds suggests that honokiol is inhibiting an activity that is common to multiple malignancies. Oncogenic transcription factor FOXM1 is one of the most overexpressed oncoproteins in human cancer. Here we found that honokiol inhibits FOXM1-mediated transcription and FOXM1 protein expression. More importantly, we found that honokiol’s inhibitory effect on FOXM1 is a result of binding of honokiol to FOXM1. This binding is specific to honokiol, a dimerized allylphenol, and was not observed in compounds that either were monomeric allylphenols or un-substituted dihydroxy phenols. This indicates that both substitution and dimerization of allylphenols are required for physical interaction with FOXM1. We thus demonstrate a novel and specific mechanism for FOXM1 inhibition by honokiol, which partially may explain its anticancer activity in cancer cells.

The quantum yield for formation of this doubly charge-separated photoproduct is 0.5%. In acidic oxygen-free solution, the reduction product is a stable dithiol. Under steady-state photoirradiation, our heptad catalyzes the two-electron reduction of an aliphatic disulfide via thiolate-disulfide interchange. Exploitation of potential inversion for the reversible light-driven accumulation of redox equivalents in artificial systems is unprecedented and the use of such a charge-accumulated state for multielectron photoredox catalysis represents an important proof-of-concept.
