Dissolve 0.486 g (1.86 mmol) of 5-bromo-2,3-dimethoxybenzoic acid in 10 ml of dry toluene, add 0.39 ml of thionyl chloride, and heat for 1 hour under the protection of argon at 60 ¡ã C., evaporate dry,After addition of 10ml of anhydrous DCM, 0.286g of (S) -2-aminomethyl-1-ethylpyrrole was added dropwise to the ice-water bath. After the reaction was stirred at room temperature for 8 hours, it was diluted with 50ml of DCM, washed with saturated aqueous NaHCO3 Washed three times,The organic layer was dried over Na2SO4, filtered and evaporated to give a yellowish oil which was separated on a medium pressure silica gel column with mobile phase petroleum ether: ethyl acetate 2: 1. The product fractions were collected and evaporated to give 1.628 g of a slightly yellow oil 87.5percent.

General procedure: Benzoic acid derivatives (1 mmol) and heterogeneous catalyst (10 mg) were mixed for 5 min in 1 mL of anhydrous toluene. Then, the amine (1.2 mmol) was added and the mixture was reacted under ultrasound for about 15-60 min at room temperature. After completion of the reaction (monitored by TLC), the catalyst was separated through filtration and the solvent was removed in vacuo. The obtained residue was dissolved in chloroform (10 ml) and washed with 10% NaHCO3 (10 ml) and HCl (1 M, 10 ml). The organic layer was extracted and dried over Na2SO4 and concentrated under reduced pressure to afford the amide, which was purified by recrystallization or column chromatography.

To a mixture of cyanuric chloride (0.368 g, 2 mmol) inCH3CN at approximately-10to-20 C was added3-fluoro-p-anisidine (0.28 g, 2 mmol) inCH3CN followed by the additionof N, N diisopropylethylamine (DIEA) (0.35 mL, 2 mmol) and stirred for an hour. The reaction mixture was then allowed to reach room temperature for an hour. The second step was continued without further purification. Cycloheptylamine (0.25 mL, 2 mmol) and DIEA (0.35 mL, 2 mmol) were added and the reaction mixture was stirred overnight at rt. The third step was also preceded without any further purification. S-(-)-2-aminomethyl-N-ethyl pyrrolidine (0.29 mL, 2 mmol) and DIEA (0.35 mL, 2 mmol) were added and the reaction mixture was refluxed overnight. The reaction mixture was diluted with ethyl acetate and washed with brine. The organic layer was separated and dried over potassium carbonate, filtered, and concentrated under reduced pressure affording 0.920 g crude material. The crude material was purified by column chromatography to yield a white solid 139 (0.550 g,60%), mp75-77 C ; HPLC: Inertsil ODS-3V C18,40 : 30: 30[KH2P04(O. 01M, pH 3.2) :CH30H :CH3CN], 264 nm, Rt 7.9 min, 95.9% purity; MS (ESI):rnlz 458(M+H, 100).

First, 4-benzyloxy-3,5-dimethoxybenzoic acid chloride represented by Formula 85 and [(2S)-1-ethylpyrrolidin-2-yl]methanamine represented by Formula 103 were prepared in 0.2 M solution, respectively, using a toluene solvent. An amine solution (1.2 mL, 0.240 mmol) entered 10 mL vial and a sodium carbonate solution (0.4 mL, 0.200 mmol) was added thereto. Thereafter, a benzoic acid chloride solution (1.0 mL, 0.200 mmol) entered the vial, vigorously shaken and agitated at room temperature for 18 hours. After terminating the reaction, an organic layer was moved to a cartridge tube (6 mL, benzenesulfonic acid, 904030-WJ, UCT) using ethyl acetate. Impurities were removed using methanol (15 mL) while separation and purification were conducted using an elute solution (ethyl acetate/methanol/triethylamine, 20:2:1, v/v/v), resulting in a benzamide compound represented by Formula 13 as a desired product (WZ-014; 66 mg, 83percent yield). [0114] Analysis data of the produced benzamide compound is provided as follows. 1H-NMR (MeOD-d4) d 1.22(t, 3H), 1.73(m, 1H), 1.80(m, 2H), 1.98(m, 1H), 2,30(m, 1H), 2.41(m, 1H), 2.76(m, 1H), 3.02(m, 1H), 3.28(m, 2H), 3.65(dd, 1H), 3.88(s, 6H), 5.02(s, 2H), 7.17?7.46(m, 7H).

To a mixture of cyanuric chloride (0.368 g, 2 mmol) in CH3CN at approximately -10 to -20 C. was added 3-fluoro-p-anisidine (0.28 g, 2 mmol) in CH3CN followed by the addition of N,N-diisopropylethylamine (DIEA) (0.35 mL, 2 mmol) and stirred for an hour. The reaction mixture was then allowed to reach room temperature for an hour. The second step was continued without further purification. The reaction mixture was diluted with ethyl acetate and washed with brine. The organic layer was separated and dried over potassium carbonate, filtered, and concentrated under reduced pressure affording 0.920 g crude material. The crude material was purified by column chromatography to yield a white solid 139 (0.550 g, 60%), mp 75-77 C.; HPLC: Inertsil ODS-3V C18, 40:30:30 [KH2PO4 (0.01M, pH 3.2):CH3OH:CH3CN], 264 nm, Rt 7.9 min, 95.9% purity; MS (ESI): m/z 458 (M+H, 100).

Then,S- (-)-2-aminomethyl-N-ethyl pyrrolidine (0.29 mL,2 mmol) and DIEA (0.35 mL, 2 mmol) were added and the reaction mixture was refluxed overnight. The reaction mixture was diluted with ethyl acetate and washed with brine. The organic layer was separated and dried over sodium sulfate, filtered, and concentrated under reduced pressure. The crude material was purified by column chromatography eluting with 96: 3: 1 methylene chloride: methanol:conc. ammonium hydroxide to yield 143, (0.300 g, 33percent) mp53-55 C ; HPLC: Inertsil ODS-3VC18, 40:30 : 30 [KH2P04 (0.01 M, pH 3. 2) : CH30H: CH3CN], 264 nm,Rt 6.9 min, 94.1percent purity; MS (ESI):m/z 449 (M+H, 100), 381 (1.2),353 (16.2), 226 (19.9), 225 (54.3), 212 (20.5), 177 (18. 3), 164 (9.6).

0.414 g (1 mmol) of compound 4a prepared in Preparation 9 was dissolved in 10 ml of anhydrous toluene, 0.39 ml of thionyl chloride was added, and the mixture was heated at 60 ¡ã C. under argon for 1 hour and then evaporated to dryness. 10 ml of anhydrous DCM was added,(S) -2-aminomethyl-1-ethylpyrrole 0.154g under ice-water bath, the reaction was stirred at room temperature for 8 hours, diluted with 50ml of EtOAc,Washed with saturated aqueous solution of NaHCO3, saturated NaCl three times, the organic layer was dried over Na2SO4, filtered and evaporated to give a yellowish oil,The residue was separated by a medium pressure silica gel column and the mobile phase petroleum ether: ethyl acetate 1: 1 (2percent TEA (triethylamine)). The product fractions were collected and evaporated to give 0.396 g as a yellowish oil, yield 75.6percent.

In a glovebox under argon, to chamber 1 of two- chamber system S2 was added Pd(dba)2 (19.9 mg, 0.0347 mmol), PPh3 (18.2 mg, 0.0693 mmol), 4,6-dichloro-2-iodo-3-methoxyphenol 37 (221 mg, 0.693 mmol), THF (3 ml_), (S)-(l-ethylpyrrolidin-2-yl)methanamine (193 muIota_, 1.39 mmol), TEA (194 muIota_, 1.39 mmol). The chamber was sealed with a screwcap fitted with a Teflon.(R). seal. In a glovebox under argon, to chamber 2 of two-chamber system S2 was added Mo(CO)6 (183 mg, 0.693 mmol), THF (3 ml_) and pyridine (280 muIota_, 3.47 mmol) in that order. The chamber was sealed with a screwcap fitted with a Teflon.(R). seal. The loaded two-chamber system was heated to 70 ¡ãC for 19 hours. The crude reaction mixture was evaporated on silica gel and the title compound 38 was obtained after flash chromatography (5percent MeOH in CH2CI2 as eluent) as brown oil (153.7 mg, 0.443 mmol, 64percent from 37).

In a 100 mL eggplant bottle, 1.22 g of benzaldehyde, 30 mL of anhydrous ethanol,(S) -1-ethyl-2-aminomethyltetrahydropyrroline, and the mixture was heated under reflux for 24 hours.Add 0.76 g of sodium borohydride, stir for 3 hours, cool to room temperature, pour into water,The organic phase was extracted with methyl chloride and dried over anhydrous magnesium sulfate to remove the solvent to give a pale yellow viscous liquid.Adding 30 mL of absolute ethanol, 0.6 g of paraformaldehyde,2-trityl-4-di-tert-butylphenol, and the mixture was heated under reflux for 12 hours.The crude product was chromatographed on silica gel to afford L4 (3.96 g, 68.2percent) as a white solid.
