The preparation of new chiral amino alcohols that possess a flexible biphenylazepine moiety and can work as tropos catalysts, is presented. In these compounds, the sign of the induced axial chirality depends on the nature of stereogenic centers near the flexible fragment and can determine the stereochemical outcome in asymmetric reactions mediated by them. The efficiency of these new ligands has been explored in the asymmetric addition of arylzinc species prepared in situ to aromatic aldehydes. Among them, amino alcohol (1R,2S)-4 gave the best results and afforded optically active diarymethanols, which are precursors of well-known chiral drugs, in high yields and with enantioselectivity up to 96 %.

The enantioseparation of three hydroxyphenylpropionic acid isomers via diastereomeric salt formation with (1R,2S)-2-amino-1,2-diphenylethanol has been demonstrated. The racemates of all three acid isomers were successfully separated with high efficiency (0.56?0.84) after single crystallization. For 2-hydroxy-3-phenylpropionic acid 4, the configuration of the less-soluble salt was controlled by the crystallization solvent: the (R)-4 salt was crystallized from water, while 2-propanol afforded the (S)-4 salt. The chiral recognition mechanism of the three acids was discussed based on the crystal structures of the diastereomeric salts.

Polycyclic borazines with three adjacent 1,3,2-oxazaborolidine rings were efficiently synthesized by the reaction of 1,2-aminoalcohols or 1,2-aminophenol with borane at room temperature and subsequent cyclization of the intermediate above 120 C. The X-ray structure of borazine derived from norephedrine indicates an almost planar central aromatic borazine structure condensed with the oxazaborolidine rings and the phenyl and methyl groups at one side of the plane.

Chirality switching was induced by solvents in the enantioseparation of 2-hydroxy-4-phenylbutyric acid (HPBA) via diastereomeric salt formation with an enantiopure aminoalcohol. The (S)-salt was crystallized from butanol solutions and the (R)-salt was obtained from aqueous solutions. It was found from crystallographic analysis of the salts that solvent inclusion changed the solubility of both salts. In particular, the (R)-salt afforded multiple pseudo-polymorphic hydrated crystals. Successive crystallization from two different solvents efficiently afforded both pure diastereomeric salts.

Achieving enzyme-like catalytic activity and stereoselectivity without the typically high substrate specificity of enzymes is a challenge in the development of artificial catalysts for asymmetric synthesis. Polyfunctional catalysts are considered to be a promising tool for achieving excellent catalytic efficiency. A polyfunctional catalyst system was developed, which incorporates two Lewis acidic/Br°nsted basic cobalt centers in combination with triazolium moieties that are crucial for high reactivity and excellent stereoselectivity in the direct 1,4-addition of oxindoles to maleimides. The catalyst is assembled through click chemistry and is readily recyclable through precipitation by making use of its charges. Kinetic studies support a cooperative mode of action. Diastereodivergency is achievable with either Boc-protected or unprotected maleimide.

A new titanium(IV) complex has been developed for the effective enantioselective alkynylation of phenylacetylene addition to aldehydes. The titanium(IV) complex was readily prepared in situ from (R)-C-(7,7-dimethyl-2- oxo-bicyclo[2.2.1]hept-1- yl)-(1R,2S)-N-(2-hydroxy-1,2-diphenyl-ethyl)- methanesulfonamide (1h) and tetraisopropyl titanate [Ti(i-OPr)4]. A variety of aromatic aldehydes and alpha,beta-unsaturated aldehydes were found to be suitable substrates in the presence of the camphor sulfonylated amino alcohol titanium(IV) complex [10 mol% 1h, 40mol% Ti(i-OPr)4]. The desired propargylic alcohols were afforded with high isolated yields (up to 90%) and moderate enantioselectivities (up to 65% ee) under mild conditions. Copyright

Herein we describe a highly chemoselective A3-coupling/annulation of amino alcohols, formaldehyde, two kinds of aldehydes and alkynes, catalyzed by copper(II). This cascade reaction, employing readily available materials, provides a new and highly effective access to chiral N-propargyl oxazolidines with good diastereoselectivity (up to >20:1). In the case of ortho-substituted aromatic aldehydes, an intriguing steric effect is observed: a bulky group exhibits a remarkably adverse effect on the diastereoselectivity for the formation of the title molecule.
