Peptide nucleic acid (PNA) is an achiral nucleic acid mimic with a backbone consisting of partly flexible aminoethyl glycine units. In contrast, incorporation of the (R.R) isomer- resulted in a drastic decrease in the stability of tile PNA DNA (or RNA) complex (DeltaT(m)= 8C per (R,R) unit). In PNA PNA duplexes, however, the (R,R)- and (S,S)-cyclohexyl residues only exerted a minor effect on the stability, and the complexes formed with the two isomers are of opposite handedness, as evidenced from circular dichroism spectroscopy. In some cases the introduction era single (S,S) residue in a PNA 15-mer improves its sequence specificity for DNA or RNA. From the thermal stabilities and molecular modeling based on the solution structure of a PNA DNA duplex determined by N MR techniques, we conclude that the right-handed helix can accommodate the (S,S) isomer more easily than the (R,R) isomer. Thermodynamic measurements of DeltaH and AS upon PNA DNA duplex formation show that the introduction of an (S,S)-cyclohexyl unit in the PNA does indeed decrease the entropy loss, indicating a more conformationally constrained structure. However, the more favorable entropic contribution is balanced by a reduced enthalpic gain, indicating that the structure constrained by the cyclohexyl group is not so well suited for DNA hybridization.

The first example of a bifunctional organocatalyst assembled through dynamic covalent chemistry (DCC) is described. The catalyst is based on reversible imine chemistry and can catalyze the Morita-Baylis-Hillman (MBH) reaction of enones with aldehydes or N-tosyl imines. Furthermore, these dynamic catalysts were shown to be optimizable through a systemic screening approach, in which large mixtures of catalyst structures were generated, and the optimal catalyst could be directly identified by using dynamic deconvolution. This strategy allowed one-pot synthesis and in situ evaluation of several potential catalysts without the need to separate, characterize, and purify each individual structure. The systems were furthermore shown to catalyze and re-equilibrate their own formation through a previously unknown thiourea-catalyzed transimination process.

By using a thermochemical cycle, the experimentally inaccessible one-electron reduction potentials for 4,4?-HO-Ph-Ph-O and 2,2?–O-Ph-Ph-O were calculated to be 0.64 and 0.78 V vs NHE, respectively. From the redox and acidity data we also estimated the O-H bond dissociation enthalpies for 4,4?-HO-Ph-Ph-OH, 4,4?–O-Ph-Ph-OH, 2,2?-HO-Ph-Ph-OH, and 2,2?–O-Ph-Ph-OH to be 349, 330, 372, and 385 kJ mol-1, respectively. The results are discussed in light of previously established substituent effects on the thermochemistry of phenols and phenoxyl radicals.

Chiral diphosphite ligands L1-L3 were prepared by the reaction of (1S,5S,6R)-(cis,trans)-spiro[4.4]nonane-1,6-diol with chlorophosphites. These ligands were tested in the rhodium catalyzed hydroformylation of styrene and enantioselectivities up to 69% were achieved. Simple mixed anhydrides are known to pose synthetic difficulties relating to their thermal lability and ways to stabilise such mixed anhydride systems by relying on either electronic or steric effects were therefore explored. Thus, a series of acyloxyphosphines and acylphosphites derived from either propanoic acid or phenylacetic acid were prepared and their in solution stability assessed. These compounds were, where stability allowed, fully characterised using standard analytical techniques. NMR studies, in particular, unearthed interesting coupling behaviour for a number of the acyloxyphosphines and acylphosphites as well as their rearrangement products which could be linked to their chiral nature. Furthermore, the crystal structures for three of the prepared mixed anhydrides were determined using X-ray crystallography and are reported herein.
