Axially-unfixed 2,2 ‘-bipyridine-based chiral catalysts were synthesized using enantiopure amino acids as chiral sources, which were successfully used in asymmetric aldol reactions ofp-nitrobenzaldehyde with cyclohexanone, while the bulkier aldehyde derivatives (2-naphthaldehyde, 9-anthracenecarboxaldehyde, and 1-pyrenecarboxaldehyde) were selected to further elucidate the catalytic properties. Particularly, the influences of the bipyridine-proline chiral structures and the polarities of used solvents (petroleum ether, toluene, CH2Cl2, ethanol, DMF, DMSO, and water) on the catalytic performance were investigated. The results indicated that the structure of bipyridine catalysts, the molecular volume of aldehydes, and the polarity of solvents have significant effects on the catalytic activities, in which, the smaller steric effects were conducive to the improvement of the yields and stereoselectivities for asymmetric aldol reaction, along with the increased polarity of used solvents and the decreased molecular volume of aldehydes. Meanwhile, the chemical identity of all compounds was confirmed by(1)H-NMR,C-13-NMR, HRMS, and HPLC analysis.

Asymmetric synthesis of mechanically planar chiral rotaxanes and topologically chiral catenanes has been a long-standing challenge in organic synthesis. Recently, an excellent strategy was developed based on diastereomeric synthesis of rotaxanes and catenanes with mechanical chirality followed by removal of the chiral auxiliary. On the other hand, its enantioselective approach has been quite limited. Here, we report enantioselective preparation of mechanically planar chiral rotaxanes by kinetic resolution of the racemates via remote asymmetric acylation of a hydroxy group in the axis component, which provides an unreacted enantiomer in up to>99.9% ee in 29% yield (the theoretical maximum yield of kinetic resolution of racemate is 50%). While the rotaxane molecules are expected to have conformational complexity, our original catalysts enabled to discriminate the mechanical chirality of the rotaxanes efficiently with the selectivity factors in up to 16. Since the discovery of mechanically planar chiral rotaxanes and topologically chiral catenanes, their asymmetric synthesis has been a long-standing challenge. Here, the authors report enantioselective preparation of mechanically planar chiral rotaxanes with up to 99.9% ee in 29% yield.

Biaryl atropisomers are key structural components in chiral ligands, chiral functional materials, natural products, and bioactive compounds, and their asymmetric syntheses have been reported by many groups. In contrast, although the scientific community has long been aware of atropisomers due to rotational restriction around N-C bonds, they have attracted scant attention and have remained an unexplored research area. In particular, their catalytic asymmetric synthesis and the synthetic applications were unknown until recently. This Account describes studies conducted by our group on the catalytic enantioselective syntheses of N-C axially chiral compounds and their applications in asymmetric reactions. In the presence of a chiral Pd catalyst, the reactions of achiral secondary ortho-tert-butylanilides with 4-iodonitrobenzene proceeded in a highly enantioselective manner (up to 96% ee), affording N-C axially chiral N-arylated ortho-tert-butylanilides in good yields. The application of the present chiral Pd-catalyzed N-arylation reaction to an intramolecular version gave N-C axially chiral lactams with high optical purity (up to 98% ee). These reactions were the first highly enantioselective syntheses of N-C axially chiral compounds with a chiral catalyst. Since the publication of these reactions, N-C axially chiral compounds have been widely accepted as new target molecules for catalytic asymmetric reactions. Furthermore, chiral-Pd-catalyzed intramolecular N-arylations were applied to the enantioselective syntheses of N-C axially chiral quinoline-4-one and phenanthridin-6-one derivatives. We also succeeded in the enantioselective syntheses of various N-C axially chiral compounds using other chiral Pd-catalyzed reactions. That is, optically active N-C axially chiral N-(2-tert-butylphenyl)indoles, 3-(2-bromophenyl)quinazolin-4-ones, and N-(2-tert-butylphenypsulfonamides were obtained through chiral Pd-catalyzed 5-endo-hydroaminocyclization, monohydrodebromination (reductive asymmetric desymmetrization), and Tsuji-Trost N-allylation, respectively. The study of the catalytic asymmetric synthesis of axially chiral indoles has contributed to the development of not only N-C axially chiral chemistry but also the chemistry of axially chiral indoles. Subsequently, the catalytic asymmetric syntheses of various indole derivatives bearing a C-C chiral axis as well as an N-C chiral axis have been reported by many groups. Moreover, axially chiral quinazlolin-4-one derivatives, which were obtained through chiral Pd-catalyzed asymmetric desymmetrization, are pharmaceutically attractive compounds; for example, 2-methyl-3-(2-bromophenyl)quinazolin-4-one product is a mebroqualone possessing GABA agonist activity. Most of the N-C axially chiral products have satisfactory rotational stability for synthetic applications, and their synthetic utility was also demonstrated through application to chiral enolate chemistry. That is, the reaction of various alkyl halides with the enolate prepared from the optically active anilide, lactam, and quinazolinone products proceeded with high diastereoselectivity by asymmetric induction due to the N-C axial chirality. At the present time, N-C axially chiral chemistry has become a popular research area, especially in synthetic organic chemistry, and original papers on the catalytic asymmetric syntheses of various N-C axially chiral compounds and their synthetic applications have been published.

X-Ray and NMR Studies on Host-Guest Inclusion Complex Formation between Crown Ethers and Pyridinium Compounds

Inclusion complex formation between benzene-substituted crown ethers and electron-deficient pyridinium ions was studied by crystallographic and NMR methods. The major attractive host-guest interactions in these complexes are face-to-face aromatic-aromatic and cation-? interactions. In addition, the crystal structures show that hydrogen bonding influences the complexation of cations.Individual studies of the binding strength as a function of host, guest, and solvent were carried out.Four pyridinium guests were prepared for the investigation.Fast atom bombardment (FAB) mass spectrometry was used to determine the stoichiometry of the complexes. The stability constants were measured by 1H NMR and the structures of the complexes in acetonitrile are discussed.X-ray crystal structures were determined for complexes of dibenzo-18-crown-6 with pyridinium tetrafluoroborate (2B18C6-PyBF4) and dibenzo-18-crown-6 with 1-aminopyridinium tetrafluoroborate (2B18C6-1-NH2PyBF4). – Keywords: crown compounds; cations; host-guest chemistry; pi interactions

UV and IR spectroscopic studies of cold alkali metal ion-crown ether complexes in the gas phase

We report UV photodissociation (UVPD) and IR-UV double-resonance spectra of dibenzo-18-crown-6 (DB18C6) complexes with alkali metal ions (Li+, Na+, K+, Rb+, and Cs+) in a cold, 22-pole ion trap. All the complexes show a number of vibronically resolved UV bands in the 36 000-38 000 cm-1 region. The Li+ and Na+ complexes each exhibit two stable conformations in the cold ion trap (as verified by IR-UV double resonance), whereas the K+, Rb +, and Cs+ complexes exist in a single conformation. We analyze the structure of the conformers with the aid of density functional theory (DFT) calculations. In the Li+ and Na+ complexes, DB18C6 distorts the ether ring to fit the cavity size to the small diameter of Li+ and Na+. In the complexes with K+, Rb +, and Cs+, DB18C6 adopts a boat-type (C2v) open conformation. The K+ ion is captured in the cavity of the open conformer thanks to the optimum matching between the cavity size and the ion diameter. The Rb+ and Cs+ ions sit on top of the ether ring because they are too large to enter the cavity of the open conformer. According to time-dependent DFT calculations, complexes that are highly distorted to hold metal ions open the ether ring upon S1-S 0 excitation, and this is confirmed by extensive low-frequency progressions in the UVPD spectra.

The design of catalysts for asymmetric propargylations remains a challenging task, with only a handful of methods providing access to enantioenriched homopropargylic alcohols. In this work, guided by previously reported computational predictions, a set of atropisomeric bipyridine N,N’-dioxides was tested as Lewis base catalysts for the asymmetric propargylation of aldehydes with trichloroallenylsilane. The catalysts are easily prepared in four simple steps starting from readily available methyl ketones. Aryl-substituted derivatives proved to be highly active and showed a high level of enantiocontrol even at 1 mol% loading. The reaction scope includes a wide range of aromatic, heteroaromatic, and unsaturated aldehydes. New computations confirm that the key stereodetermining transition state structures for the synthesized catalysts are similar to those previously reported for the model structure.
